If you have a HDTV (which is needed for playing Minecraft in splitscreen-mode), then just:

1. Start a singleplayer-game with the first controller.

2. Log in with a profile on the second controller.

3. Press the "Start" button on the second controller.

4. Congrats, now you can play splitscreen.

The console will need to be hooked up to an HDTV with a minimum output of 720p in order for split screen to work.  Click the link to find out how to set that up on your console.
