Net Framework Initialization Error

I am having a persistant problem with a 'Framework Initialization error' saying that windows\microsoft.net\Framework\v2.0.50727\mscorwks.dll could not be loaded.  This occurs when i try and load a particular programme. I cannot reload the programme as i get a error message then. Can I get beyond this?

First of all ensure that your filesystem state is ok, by using this command:
chkdsk %SystemDrive% /f

Then you can try to reinstall .Net Framework v.2.0. Download it from Microsoft:
For x86 system:
http://www.microsoft.com/download/en/details.aspx?id=19
For x64 system:
http://www.microsoft.com/download/en/details.aspx?id=6523

After that you can download .Net Framework v.2.0 Service pack 2 from here:
http://www.microsoft.com/download/en/details.aspx?id=1639

After that you will need to install a .Net update from here, for your OS:
http://support.microsoft.com/kb/959209
