I have built a Intel system for a customer and have had problem of one kind to another . The MB is a DP43TF has a core 2 DUO Processor 4GB Mem and Windows 7 Pro 64 bit OS. Ran it for three days the Video Card went out, replaced it Did a complete reinstall of OS then ran it for two days with no problems. I shut it down for Approx. 6 Hrs and then turned the unit back on. I got 3 Beeps and the system would not start I used the reset switch again the same thing. So I used the power switch to shutdown and restart. The system start perfect and will run with no problems for 24 hrs or so If I do a restart it performs just fine. If I shut the unit off for several hours the restart I start all over again with the three beeps. What could cause this? Will this be a MB problem?

Most likely is a Hardware issue.
Refer to the Motherboard manual, and look for the three beeps on the beep/error codes section, to find the probable cause.
PS: From that model's Manual:
Table 15. Beep Codes
Beep Description
3 No memory
Siren Processor overheat (on reboot)
Check the RAM modules.
