Windows 10 updates do have a habit of changing the requirement for the Fn key when using the function keys (F1 - F12)
If you press Alt + Fn + F4, does that result in the original action?
If so, try pressing Fn + Exc to turn off the need for that Fn key
Then try pressing Fn + F4
If none of the above works for you, what is the Make and exact Model No. of your PC - that will be printed on a sticker on the bottom casing . ..

To check if that update has corrupted your keyboard setup, please follow these steps:
Open the Settings App
Go to Update & Security - Troubleshoot
Run the Keyboard troubleshooter
If that does not resolve this for you
Open Device Manager
Expand the Keyboards section
Right click your keyboard and choose 'Uninstall'
Then restart your PC
